100,000+ accounts? Wow!

No experience with that, but we use two webmail apps 
here:

WebMail v3.51 - http://www.ihub.com/products/WebMail

Mirapoint - http://www.mirapoint.com

For the last couple of days, the first product has been 
really slow dealing with only 300+ users who seem to 
be doing a lot of housecleaning (deleting large numbers 
of mail messages, for example). I wouldn't recommend 
it for 100,000+ users!

Don't know how Mirapoint would hold up.

I suspect you'd have to come up with some sort of 
webmail and mail server farm to provide acceptable 
response for that many accounts. I think that's how 
places like Hotmail and such do it.
